What is dC(Bz)?
dC(Bz) refers to N4-Benzoyl-2′-deoxycytidine, commonly called N4-benzoyl-protected deoxycytidine.
- dC = 2′-deoxycytidine (one of the four DNA nucleobases)
- (Bz) = Benzoyl, a base-protecting group
Why is Bz protection needed for dC?
Cytosine (C) has a reactive exocyclic amino group (N4-NH₂), which, if left unprotected, can cause:
- Increased side reactions
- Reduced coupling efficiency
Using Benzoyl (Bz) protection safeguards the N4-amino group, allowing DNA synthesis to proceed more stably, with stronger acid and oxidation resistance.
Applications of dC(Bz)-CPG
dC(Bz)-CPG is most commonly used to synthesize DNA oligonucleotides with a 3′-end cytosine. Typical applications include:
- PCR / qPCR primers (30–60 nt)
- DNA probes (20–40 nt)
- Antisense oligonucleotides (ASO, 15–25 nt)
- gRNA template DNA and other gene-editing related DNA
It is particularly recommended when the DNA sequence is >50 nt, or when dC(Ac) fails or shows instability. For short oligos (<20 nt), dC(Bz)-CPG is rarely required.
