Proprietary Hybrid Architecture:

Controlled Pore Glass (CPG) Reinforced with Functionalized Polymer Coating

Poresyn Solutions has pioneered and patented a breakthrough solid support technology engineered specifically for modern oligonucleotide synthesis—with specialized optimization for complex, extra-long DNA and RNA sequences.

By covalently combining rigid Controlled Pore Glass (CPG) microparticles with an engineered styrene/nitrogen-heterocycle copolymer coating (with optional long-chain alkyl ether linkers), this hybrid carrier delivers unprecedented loading capacities and step-wise coupling efficiencies while completely bypassing the historic pitfalls of traditional solid supports.

The Core Bottlenecks of Conventional Solid Supports

For decades, solid-phase oligonucleotide elongation has relied on two conventional formats, forcing scientists and manufacturers into a difficult compromise:

Conventional Polystyrene (PS) Resins

Advantage: High loading capacity. Fatal Limitations: Unsuitable for longer chains (>25–30 nt) due to severe steric crowding. Furthermore, low-crosslinked PS resins swell by 300% to 600% in common organic synthesis solvents (e.g., DMF, DCM, acetonitrile). This drastic swelling creates erratic column backpressure, restricted mass transfer, flow resistance, and massive chemical consumption.

Functionalized Controlled Pore Glass (CPG)

Advantage: Rigid, dimensionally stable, non-swelling skeleton with defined pore channels, historically preferred for medium-to-long fragments. Fatal Limitations: Severe trade-off between pore size and loading density. Large pores (1000 Å–2000 Å) suffer from drastically diminished surface areas, yielding poor functional loading (<15–30 μmol/g). Additionally, native silica matrices are prone to chemical etching under basic deprotection conditions, leaching silicon contaminants into the API and driving up purification costs.

PoreSyn’s Patented Innovation: Organic/Inorganic Hybridization

PoreSyn’s patented platform directly addresses the fundamental limitations of conventional CPG and polymer-based solid supports.

By applying an ultra-thin, nanoscale layer of a specialized styrene/nitrogen-containing heterocyclic copolymer onto high-purity CPG microparticles—with the coating thickness precisely controlled at a 1:200 to 1:250 thickness-to-pore-diameter ratio—PoreSyn creates a unique organic/inorganic hybrid architecture.

The result combines the functional density of organic polymers with the mechanical rigidity and porous structure of glass.

01 / LOADING

Hyper-Loading Capacities

Because elongation takes place on the crosslinked, three-dimensional functional coating, PoreSyn carriers routinely deliver 2× to 4× the effective loading capacity of native CPG across different pore sizes.

110–140 μmol/g · 1000 Å
60–90 μmol/g · 2000 Å
02 / COUPLING

Accelerated Coupling via Catalytic Heterocycles

Nitrogen-containing heterocyclic rings covalently integrated into the polymer backbone interact directly with incoming phosphoramidite monomers.

This interaction creates a highly reactive, intramolecular-like intermediate that promotes efficient stepwise coupling and helps suppress n−1 and n−2 truncation impurities.

03 / LINKER

Optimized Microenvironment with LCAA Units

PoreSyn’s patented Long-Chain Alkyl Ether and Amino (LCAA) linker systems provide a spatial cushion between the growing oligonucleotide chain and the solid surface.

By projecting the growing sequence away from the CPG surface, the linker architecture helps reduce steric congestion and improves reagent accessibility during complex and extended synthesis.

04 / FLUIDICS

100% Non-Swelling Fluidic Performance

Unlike bulk polymer resins, PoreSyn’s ultra-thin organic film is structurally anchored to the porous silica skeleton.

The hybrid carrier exhibits essentially zero volumetric swelling, preserving the original pore architecture during synthesis.

  • Stable fluidic behavior
  • Consistent pressure profiles
  • Reliable reagent flow
  • Reduced risk of column blockage
  • High-pressure automated synthesis compatibility
05 / PURITY

Elimination of Silicon Leaching

The dense, chemically resilient copolymer coating forms a protective layer over the internal silica surface.

This architecture helps shield the inorganic CPG substrate from direct reagent attack, reducing the risk of silicon-containing particulate contamination and supporting cleaner downstream processing.

06 / LONG-MER

Long-Mer RNA & DNA Fidelity

The combination of high loading, efficient coupling, optimized linker architecture, and stable pore structure enables continuous solid-phase assembly of long DNA and RNA sequences ranging from approximately 80 nt to 300+ nt.
